New NMR Tools for Interrogation of Chemical Reactions and Processes.

This project seeks to advance unique NMR hardware development in Edinburgh for reaction mechanism and kinetics, which allow interfacing of precision stop-flow and quench flow techniques with custom-built NMR instruments. The student will contribute towards interfacing of chemical reactors with NMR spectrometers, automation of the monitoring of reaction kinetics, and in particular customising of existing and the development of novel NMR experiments for this area including data evaluation.
Analysis of complex mixtures
We are developing multidimensional NMR methods and mass spectrometry approaches, in combination with chemical modification schemes, to investigate the most complex mixtures on earth - humic substances - heterogeneous mixtures containing thousands of organic molecules produced by the degradation of plant and animal residues. We also study other, more pleasant mixtures, like Scotch whisky.
Carbohydrate structures and protein-carbohydrate interactions
Glycosaminoglycans isolated from marine environment are complex, heterogeneous polysaccharides that represent a considerable challenge for the NMR structure determination. We investigate interactions between glycosaminoglycans and selectins – proteins involved in inflammation and metastasis progression. We also study the conformation of carbohydrates using scalar and residual dipolar coupling constants.
NMR methodology
Using our 400-800 MHz NMR spectrometers equipped with cryoprobes we are developing NMR methodologies for efficient acquisition of NMR spectra applicable across the field of chemistry and biochemistry. This includes fast NMR methods for kinetic and mechanistic studies.
Development of NMR methodology and its application to studies of complex mixtures, chemical reactions, carbohydrate structures and protein-carbohydrate interactions.
